RESUMO

The threat of bioterrorism has spurred research on the decontamination and containment of different agents. Anthrax [causative agent Bacillus anthracis (Ba)] is a disease that can lead to severe infections within human and animals, particularly when inhaled. This research investigated the use of spore-contaminated simulated runoff events into stormwater control measures (SCMs), which are designed to retain and improve the quality of runoff and may have the potential to filter and contain the spores. In this study, the effectiveness of a bioretention cell (BRC) and high flow media filter (HFMF) in Huron, Ohio, were evaluated for removal of Bacillus globigii (Bg) spores (a harmless cognate of Ba). Three 4-8 mm simulated runoff events were created for each SCM using a fire hydrant and Bg spores were injected into the runoff upstream of the SCM inlets. The BRC significantly (p < 0.001) outperformed the HFMF in reducing Bg concentrations and loads, with an average load reduction of 1.9 log (∼99% reduction) compared to 0.4 (∼60% reduction), respectively. A probable critical design factor leading to these differences was the infiltration rate of the media and subsequent retention time within the filters, which was supported by similar disparities in suspended solids reductions. Differences in spore removal may also have been due to particle size distribution of the HFMF, which was more gravelly than the bioretention cell. At 3 and 6 months after the-simulated runoff tests, soil samples taken from both SCMs, yielding detectable Bg spores within the top 15 cm of media, with increased spore concentrations where ponding occurred for longer durations during the tests. This suggests that forebays and areas near inlets may be hotspots for spore cleanup in a real-world bioterrorism incident.

PURPOSE: This study explored the potential feasibility of cell-free DNA (cfDNA) in monitoring treatment response through the measurement of chromosomal instabilities using I-scores in the context of radiation therapy (RT) for other solid tumors. MATERIALS AND METHODS: This study enrolled 23 patients treated with RT for lung, esophageal, and head and neck cancer. Serial cfDNA monitoring was performed before RT, 1 week after RT, and 1 month after RT. Low-depth whole-genome sequencing was done using Nano kit and NextSeq 500 (Illumina Inc.). To measure the extent of genome-wide copy number instability, I-score was calculated. RESULTS: Pretreatment I-score was elevated to more than 5.09 in 17 patients (73.9%). There was a significant positive correlation between the gross tumor volume and the baseline I-score (Spearman rho = 0.419, p = 0.047). The median I-scores at baseline, post-RT 1 week (P1W), and post-RT 1 month (P1M) were 5.27, 5.13, and 4.79, respectively. The I-score at P1M was significantly lower than that at baseline (p = 0.002), while the difference between baseline and P1W was not significant (p = 0.244). CONCLUSION: We have shown the feasibility of cfDNA I-score to detect minimal residual disease after RT in patients with lung cancer, esophageal cancer, and head and neck cancer. Additional studies are ongoing to optimize the measurement and analysis of I-scores to predict the radiation response in cancer patients.

This study characterized Bacillus globigii (BG) as a Bacillus anthracis Sterne (BAS) surrogate for wastewater treatment-related studies of UV inactivation, adsorption onto powdered activated carbon (PAC), and bioaerosol emission. The inactivation of BG was faster than that of BAS in DI water (pseudo first-order rate constants of 0.065 and 0.016 min-1 respectively) and in PBS solution (0.030 and 0.005 min-1 respectively). BG was also removed more quickly than BAS by PAC adsorption in DI (0.07 and 0.05 min-1 respectively) and in PBS (0.09 and 0.04 min-1 respectively). In DI, BG aggregated more (P < 0.05) than BAS when the pH was 7 or greater but there were no statistically significant differences in NaCl solution. Spore aggregation was also studied with extended Derjaguin-Landau-Verwey-Overbeek (XDLVO) models. Less than 1% of all spores were released as bioaerosols, and there was no significant difference (P > 0.05) in emission between BG and BAS. To the author's knowledge, this study is the first to demonstrate that BG is a suitable surrogate for BAS for bioaerosol emissions, but a poor surrogate for both UV inactivation and PAC adsorption. These results can be used to understand the ability of BAS to act as a surrogate for BA Ames because of its genetic and morphological similarities with BAS.

Synbiotics, including probiotics and prebiotics, are useful for patients with functional bowel disorders. However, which synbiotics are beneficial for patients with which diseases, especially those with functional diarrhea (FDr) with high fecal calprotectin levels, is currently unknown. FDr is an extension of irritable bowel syndrome with diarrhea (IBS-D). Although fewer studies have been conducted on FDr compared to IBS-D, its importance is increasing as its prevalence increases. The aim of this study was to evaluate the effects of a synbiotic containing a mixture of Lactobacillus and Bifidobacterium and its substrate, fructooligosaccharide, on bowel symptoms, fecal calprotectin levels, fecal microbiota, and safety in FDr patients with high fecal calprotectin levels. Forty patients were randomly assigned to either a synbiotic group or a placebo group. A total of 20 subjects in the synbiotic group and 19 subjects in the placebo group completed the study (8 weeks). Changes in FDr symptoms, fecal calprotectin levels, and gut microbiota were assessed during the intervention period. At 4 and 8 weeks, the number of bowel movements tended to increase in the synbiotic group, with a significant increase in the number of formed stools rather than loose stools (p < 0.05). Bowel movement satisfaction was significantly increased in the synbiotic group, but not in the placebo group. Intestinal flora analysis revealed that Lactobacillales at the order level was increased only in the synbiotic group at the end of the intervention. In contrast, at week 8 of the intervention, log-transformed fecal calprotectin levels were significantly decreased in the synbiotic group, although the change was not significantly different from that of the placebo group. These findings suggest that the intake of a multi-strain-containing synbiotic for 8 weeks could improve gut symptoms and the intestinal microenvironment of FDr patients with high fecal calprotectin levels.

Challenges exist in the wastewater treatment of dyes produced by the world's growing textiles industry. Common problems facing traditional wastewater treatments include low retention values and breaking the chemical bonds of some dye molecules, which in some cases can release byproducts that can be more harmful than the original dye. This research illustrates that track-etched polycarbonate filtration membranes with 100-nanometer diameter holes can be functionalized with azo dye direct red 80 at 1000 µM, creating a filter that can then be used to remove the entire negatively charged azo dye molecule for a 50 µM solution of the same dye, with a rejection value of 96.4 ± 1.4%, at a stable flow rate of 114 ± 5 µL/min post-functionalization. Post-functionalization, Na+ and NO3- ions had on average 17.9%, 26.0%, and 31.1% rejection for 750, 500, and 250 µM sodium nitrate solutions, respectively, at an average flow rate of 177 ± 5 µL/min. Post-functionalization, similar 50 µM azo dyes had increases in rejection from 26.3% to 53.2%. Rejection measurements were made using ultraviolet visible-light spectroscopy for dyes, and concentration meters using ion selective electrodes for Na+ and NO3- ions.

Activated carbon can be manufactured from waste coffee grounds via physical and/or chemical activation processes. However, challenges remain to quantify the differences in surface morphology between manufactured activated carbon granules and the waste coffee grounds. This paper presents a novel quantitative method to determine the quality of the physical and chemical activation processes performed in the presence of intensity inhomogeneity and identify surface characteristics of manufactured activated carbon granules and the waste coffee grounds. The spatial density was calculated by the Getis-Ord-Gi* statistic in scanning electron microscopy images. The spatial characteristics were determined by analyzing Ripley's K function and complete spatial randomness. Results show that the method introduced in this paper is capable of distinguishing between manufactured activated carbon granules and the waste coffee grounds, in terms of surface morphology.

Development of numerical models to predict stormwater-mediated transport of pathogenic spores in the environment depends on an understanding of adhesion forces that dictate detachment after rain events. Zeta potential values were measured in the laboratory for Bacillus globigii and Bacillus thuringiensis kurstaki, two common surrogates used to represent Bacillus anthracis, in synthetic baseline ultrapure water and laboratory prepared stormwater. Zeta potential curves were also determined for materials representative of urban infrastructure (concrete and asphalt). These data were used to predict the interaction energy between the spores and urban materials using Derjaguin-Landau-Verwey-Overbeek (DLVO) modeling. B. globigii and B. thuringiensis kurstaki sourced from Yakibou Inc., were found to have similar zeta potential curves, whereas spores sourced from the U.S. military's Dugway laboratory were found to diverge. In the ultrapure water, the modeling results use the laboratory data to demonstrate that the energy barriers between the spores and the urban materials were tunable through compression of the electrical double layer of the spores via changes of ionic strength and pH of the water. In the runoff water, charge neutralization dominated surface processes. The cations, metals, and natural organic matter (NOM) in the runoff water contributed to equalizing the zeta potential values for Dugway B. globigii and B. thuringiensis kurstaki, and drastically modified the surface of the concrete and asphalt. All DLVO energy curves using the runoff water were repulsive. The highest energy barrier predicted in this study was for Dugway B. globigii spores interacting with a concrete surface in runoff water, suggesting that this would be the most challenging combination to detach through water-based decontamination.

We used a DC-sputtering method to deposit the precursor (Cu3Ga/In) onto Mo with 1 um thick/soda-lime glass (SLG). We moved it onto a graphite crucible for the pre-annealing process, and the pressure of the process tube was about 10 torr without Ar gas flow. The crucible in quartz tube was heated by halogen lamp to 250 degrees C for 30 min, and then raised to 550 degrees C for 10 min under a selenium atmosphere. To complete the solar cells, a buffer layer of 50 nm CdS was then deposited by chemical bath deposition (CBD), followed by a double layer (high resistivity/low resistivity) of RF sputtered i-ZnO/Al-ZnO thin films. The Al front contacts were deposited by thermal evaporator.

This article examines the influence of three common stabilizing agents (citrate, poly(vinylpyrrolidone) (PVP), and branched poly(ethylenimine) (BPEI)) on the attachment affinity of silver nanoparticles to ceramic water filters. Citrate-stabilized silver nanoparticles were found to have the highest attachment affinity (under conditions in which the surface potential was of opposite sign to the filter). This work demonstrates that the interaction between the electrical double layers plays a critical role in the attachment of nanoparticles to flat surfaces and, in particular, that predictions of double-layer interactions are sensitive to boundary condition assumptions (constant charge vs constant potential). The experimental deposition results can be explained when using different boundary condition assumptions for different stabilizing molecules but not when the same assumption was assumed for all three types of particles. The integration of steric interactions can also explain the experimental deposition results. Particle size was demonstrated to have an effect on the predicted deposition for BPEI-stabilized particles but not for PVP.

The understanding of nano-sized particle separation processes has been limited by difficulties of nanoparticle characterization. In this study, nanoparticle tracking analysis (NTA) was deployed to evaluate the absolute particle size distributions in laboratory scale flocculation and filtration experiments with silver nanoparticles. The results from NTA were consistent with standard theories of particle destabilization and transport. Direct observations of changes in absolute particle size distributions from NTA enhance both qualitative and quantitative understanding of particle separation processes of nano-sized particles.
